Developing an immunohistochemistry test for “programmed cell death 1 ligand” (PD-L1) as a companion diagnostic for pembrolizumab

Tumors express PD-L1 to escape from immunosurveillance. Pembrolizumab blocks this escape mechanism and thus effectively treats a number of cancers. The rapid clinical development of pembrolizumab required rapid development of an immunohistochemistry assay for PD-L1. Merck developed the assay initially to determine whether or not PD-L1 is a predictive biomarker, then to enrich clinical trials, and ultimately partnered with a diagnostics company to develop the assay as a companion diagnostic. This presentation describes assay development, including the decision to exclude immune cells from scoring, the decision to use the tumor proportion score, and the derivation of the 50% cutoff.

This is primarily due to the progress of translational research, which applies the results of basic science to solving the clinical problems which clinical oncologists face everyday..............................

Dr. Sasano gave his lecture taking Q&A system during his talk.
It was a good opportunity especially for young researchers to listen to his precious talk which comes from his involvement in editing of journals. He left his message emphasizing that it is very important for them to read journals more carefully and use their logical thinking skills.
